چکیده

ساختار ژنتیکی ماهی کفال طلایی Liza aurata در تالابهای گمیشان و انزلی با استفاده از 6 جایگاه میکروستلایت (Muce55 ،Muce37 ،Muso10،Muco16 ،Muso19 ،Muso22) بررسی گردید. در مجموع ٦٠ نمونه بالغ ماهی کفال از دو منطقه تالابهای گمیشان و انزلی جمع آوری شد. تمامی پرایمرهای مورد استفاده در طی واکنش زنجیره ای پلی مراز با موفقیت تکثیر شدند و چند شکل (پلی مورف) نشان دادند که از آنها برای تعیین تمایز ژنتیکی ماهی کفال طلایی استفاده شد. میانگین آللی (Na) در جایگاه‌ها 3/5 (با دامنه 3 تا 9 آلل) و در مناطق نمونه برداری در نمونه‌های تالاب گمیشان و انزلی به ترتیب 1/5 و 5/5 بود. در نمونه های هر دو منطقه آلل‌های اختصاصی دیده شد. میانگین هتروزیگوسیتی مورد انتظار و مشاهده شده به ترتیب در نمونه‌های تالاب گمیشان 721/0 و 153/0 و انزلی 747/0 و 328/0 محاسبه شد. تمامی جایگاه‌ها در بررسی تعادل هاردی وینبرگ (H-W) خارج از تعادل بودند (001/0>P). میانگین ضریب خویشاوندی ( Fisو Fit) در 6 جایگاه میکروستلایت در هر دو جمعیت مثبت بود. میزان شاخص تمایز (Fst) و جریان ژنی (Nm) بر اساس فراوانی آللی به ترتیب 113/0 و 97/1 محاسبه شد. بر اساس تست AMOVA، شاخص‌های تمایز Fst و Rst تفاوت معنی‌داری بین جمعیت‌ها نشان دادند (01/0P≤). میزان فاصله ژنتیکی نیز نشان دهنده تمایز ژنتیکی بین جمعیت‌های مورد مطالعه بود. نتایج این بررسی نشان می‌دهد که جمعیت‌های متمایز ژنتیکی از ماهی کفال طلایی در جنوب دریای خزر، در مناطق مورد بررسی (تالابهای گمیشان و انزلی) موجود است.
